Instead of blocking unfavorable proteins, could we influence the cell to throw them away?
This is achievable through molecular glues: small-molecule drugs that act like cellular matchmakers, connecting unwanted disease-targets with an enzyme that marks the target for destruction by proteasome.

A molecular glue induces a cell to degrade an unfavorable protein.
Mechanism of Action: The molecular glue promotes or stabilizes an interaction between the target protein and an E3 ubiquitin ligase enzyme. This ligase attaches ubiquitin tags to the target protein, exposing it to proteasomal degradation.

Benefits of Targeting Degradation Using Molecular Glues
- Proteins like transcription factors, RNA-binding proteins, and scaffolding proteins even though they lack the binding pockets required for conventional inhibition.
- Degradation as opposed to inhibition does not require the molecule to continuously occupy the target. This could possibly lower the exposure threshold.
- Oral delivery is feasible for some molecular-glue degraders, as shown by established cereblon-binding drugs and newer clinical candidates.
Examples of Private Companies Exploring This Modality

Path to Positive Risk-Adjusted Net Present Value – Oncology
To achieve rNPV, we need the probability-weighted commercial cash flow to exceed the discounted cost of development and manufacturing.
Oncology is the clearest near-term proving ground because of its high unmet need, biomarker-defined disease subsets, measurable target degradation, and oral exposure that supports sustained pharmacology.
For unequivocal recognition of value, we must show a credible path to efficacy, differentiation from existing treatment options, and sufficient commercial scale (relying on CMC experts).
